· The test is based on hydrolysis of substrate (sodium hippurate) by the bacterial enzyme hippurate hydrolase and production of benzoic acid and glycine. Glycine produced by this enzymatic reaction is detected after 24 hours incubation in reaction with chromogen (ninhydrine) and the blue-purple substance is produced.
· Shelf life: 12 months.
· Microbiological diagnostic test for identification of hippurate hydrolase activity of Streptococci of group B, Campylobacter jejuni, Gardnerella vaginalis, and other microorganisms.
Brief assay procedure:
1. Prepare bacterial suspension from the pure 24 hours old culture of tested microorganism in 0.5 ml of saline solution.
2. Put substrate strip into the tube with suspension so that, the whole paper zone is dipped and mix gently.
3. Incubate at 37 °C for 24 hours.
4. Add alongside the wall of the tube approx. 0.2 ml (4 drops) of the Reagent. Do not mix.
5. Incubate at room temperature (18-24 °C) for 5-10 minutes.
6. Evaluate results.
